This study investigates the expression and phosphorylation levels of the Ee;CDKF;1 protein in various organs of leafy spurge (Euphorbia esula) across different growth stages. Total proteins were phenol-extracted from the roots, hypocotyls, and cotyledons of seedlings as well as roots, stems, leaves, meristems, and flowers of mature plants. The size marker used was a protein extracted from yeast (GF2351). The proteins were separated by SDS-PAGE, transferred to membranes, and incubated with phosphatase treatments to assess their phosphorylation status.

Total proteins were phenol-extracted from root (R), hypocotyl (H), and cotyledon (C) of seedlings and root (R), stem (S), leaf (L), meristem (M), and flower (F) of mature plants. A protein extracted from yeast GF2351 harboring pYX-At;CDKF;1 (Y) was used as a size marker for Ee;CDKF;1. These proteins were fractionated by SDS-PAGE, blotted onto membranes, and incubated with PA or PP1A.
