Isolation Scheme • Initial fractionation through solvent partition • Allows separation into fractions of differing polarities Ohia leaves Methanol extraction Crude extract Hexane-methanol partition Hexane fraction Methanol fraction CH2Cl2-water partition CH2Cl2 fraction Aqueous fraction butanol-water partition Butanol fraction Water fraction

  4. Isolation Strategy • Several Fractionation steps required • Use high volume techniques first, then high resolution • Different techniques for different polarities

  5. Butanol fraction • Countercurrent fractionation Countercurrent partition system Countercurrent separation of butanol solubles HPLC analysis of countercurrent fractions

  6. Dichloromethane Fraction • High volume separation by silica column • Followed by Prep TLC, HPLC • Yields terpenoids, C-methyl-flavanoids, lipids Dr. Swapan Pramanick with HPLC 1H NMR (d6-DMSO, 400 MHz) of betulinic acid

  7. Compounds Isolated • Several compounds isolated and characterized • Wide range of structure classes lipids terpenoids flavonoid glycosides C-methyl-flavonoids
