  2. 一、实验目的 • 掌握NORs的银染色技术,了解实验原理。 • 二、实验原理 • NORs处与rDNA转录有关的酸性蛋白可被硝酸银特异染色。

  3. 三、实验用品 • 1 .材料:小鼠骨髓 • 2 .试剂:Carnoy固定液;0.01%秋水仙素;Giemsa染液;0.4%KCl低渗液;2%柠檬酸钠; 50%硝酸银溶液;2%明胶显影液(含1ml甲酸);1/15mol/L磷酸缓冲液(pH6.8)。

  4. 四、实验步骤 • 1.制片,方法同实验六; • 2.玻片置于湿盒中,60摄氏度水浴; • 3.硝酸银4滴,明胶显影液2滴3min; • 4.流水冲洗; • 5.Giemsa复染3min; • 6 .流水冲洗; • 7.加盖玻片观察。

  5. 五、作业 • 绘制小鼠骨髓细胞中期核型图,标出Ag-NORs的位置。
