FLUORESCENCE AND QUENCHING

FLUORESCENCE AND QUENCHING. Purpose. To measure the fluorescence of quinine. To measure the quenching of this emission by chloride, bromide and iodide ions and to compare the quenching efficiency of the three halides. To determine the rate constants associated with fluorescence and quenching.

  1. FLUORESCENCE AND QUENCHING

  2. Purpose • To measure the fluorescence of quinine. • To measure the quenching of this emission by chloride, bromide and iodide ions and to compare the quenching efficiency of the three halides. • To determine the rate constants associated with fluorescence and quenching.

  3. STERN-VOLMER ANALYSIS • D + hν (350 nm)  D* absorption • D*  D + hν’ (450 nm) kf fluorescence • D* + Q  D + Q kq quenching • (I0/I) = 1 + KSV [Q] KSV = kq τ0 where I0 is found from Sample B and I values correspond to Samples C-H at various [Q] values.
