  1. If = Hypothesis, Then = Prediction Prediction= What you think the outcome of a specific experiment will be Hypothesis: Why If /then statement is NOT A HYPOTHESIS You will be marked wrong.

  2. You Should Know • Scientific Method • Science Terms • Control • Controlled Variable, • Indpendent/dependent variables • Sample size • Significance • Etc…. • Tools we have learned in lab. From onions- transformations. Big picture.

  3. Hint: We are testing Scientific reasoning and problem soloving.

  4. Transformations Terms • Biotechnology: using technology to manipulate organisms • Genetic Engineering: Manipulating genes in organisms • DNA Cloning: putting a segment of DNA in an organism and having that organism make tons of copies of the gene, gene product or DNA segment. • Transformation: Introducing DNA from one organism (usually bacteria) into another organism.

  5. Very Expensive2 stopsWill be Graded on TechniqueBe patient

  6. Restriction Enzymes Leave Sticky Ends

  7. Transformation http://www.youtube.com/watch?v=aA5fyWJh5S0

  8. How do we know it worked? • Must determine if Gene was Inserted Successfully into Plasmid • Marker Genes! • GFP • Disrupt function and it disappears • Antibiotic: tells if plasmid made it in to cell • Color/Glowing: tells if gene made it into plasmid

  9. Problem

  10. Plates • Groups of 3 or 4. • Half Plates: step 7 page 104 use half volumes • 50 instead of 100, 25 instead of 50 • Check Plates 48 hours later mark on plate cover what you found. • Leave in incubator for me to grade.
