  13. Immunosuppressive effects of rapamycin Without Costimuation Mondino A et al. Semin Immunol 2007;19:162-172

  14. Immunosuppressive effects of rapamycin

  15. Immunosuppressive effects of rapamycin • Diffrentiation into Treg cells • Changing T-cell trafficking • Suppressing diffrentiation and maturation of DCs

  16. Immunostimulatory effects of rapamycin

  17. Immunostimulatory effects of rapamycin • Increased IL-12 of mDCs treated with rapamycin • Enhanced antigen presentation by rapamycin-induced • autophagy of DCs

  18. Viral infection versus tissue transplant • Viral infection has a potent ability to activate the innate • immune system • Rapamycin might inhibit the immune response only • when combined with other therapies, or rapamycin’s • immunostimulatory effects may be masked by other • drugs

  19. Targeting mTOR to enhance vaccine immunogenicity • Using rapamycin as an adjuvant • - It can be taken orally - Rapamycin does not need to be combined with the vaccine

  20. Summary • Rapamycin has immunostimulatory effects - Memory CD8+ T-cell differentiation - Increasing IL-12 of mDCs - Enhanced antigen presentation by autophagy of DCs • Rapamycin has immunosuppressive effects - Diffrentiation into Treg cells - Changing T-cell trafficking - Suppressing diffrentiation and maturation of DCs • Rapamycin can be used as a vaccine adjuvant, but it is common immunosuppressive drug for organ transplantation. So, rapamycin has to be investigated more.
