Time-dependent Gene Expression Profile in Human Hepatic Stellate Cell Proliferation and Apoptosis

This study aims to compare gene expression patterns in normal, activated, and apoptotic human hepatic stellate cell lines to identify critical genes involved in hepatic fibrosis. The hTERT cell line, PDGF-BB, and curcumin are used to induce proliferation and apoptosis. mRNA extraction, microarray hybridization, and gene expression confirmation by RT-PCR are performed.

Introduction • Hepatic fibrosis is one of the common disease in Koreans. • Preventing initial liver fibrogenesis may contribute to hepatic fibrosis therapies. • It is important to discover the mechanisms related to the proliferation and apoptosis of hepatic stellate cell line which has the critical role in liver fibrogenesis.

  3. AIM • To compare gene expression patterns time dependently between normal, activated and apoptotic human hepatic stellate cell line • To identify critical genes involved in hepatic fibrosis

  4. Materials & Method • hTERT cell line : immortalized activated human hepatic stellate cell line • PDGF-BB : Platelet Derived Growth Factor-BB ; a potent growth factor • Curcumin : a phytochemical compound known to induce apoptosis

  5. PDGF-BB and curcumin treatment on hTERT cell line • mRNA extraction of proliferation and apoptosis induced hTERT cell line • Microarray hybridization

  6. Data acquisition • Spot preprocessing, quality inspection, normalization, clustering & classification, differentially expressed gene finding & biological validation • Gene expression confirmation by RT-PCR
