SNARES (20 types) 2. Rabs ( GTPases targeting) • SNARE proteins -central role; i) specificity, ii) bringing the fusion (vesicles with the membrane in target.) Endocytic routes. • Exist in supportive sets- 1. v-SNAREs i.e vesicle membrane SNAREs • 2. t-SNAREs i.e target membrane SNAREs. • T-SNARE (trans), clasp 2 membranes together.
STRUCTURE OF SNARE SNARE motif (cytosolic domain of SNARE segment) comprised of ; • Heptadrepeats (coiled- structures); 60-70 amino acids. • Tightly reversible assembly by V- and t-SNAREs – c/a "trans"-SNARE complexes (formed of 4 -helix bundles). • 3 SNAREs (In synaptic vesicles) : • 1. syntaxin 1 (one helix) • 2. SNAP-25 (2 helix) • 3. Synaptobrevin (VAMP) - ( 1 helix)
SNAREs that're interacting, are separated apart before they can re-enact again through the action of NSF. • Rab proteins - facilitate vesicle docking rate & its regulation, matching of v-SNAREs and t-SNAREs

SNAP ( SOLUBLE NSF ATTACHTMENT PROTEINS) • Cytosolic adaptor proteins • Intricated in intracellular transport during membrane vesicular fusion &exocytosis. • Event of vesicle fusion- Assembly and de-assembly. • SNAP + SNARE + NSF = 20S complex. • SNAPs receptor proteins- syntaxin 1, • SNAP-25 (synaptosome associated protein, 25kDa), • and VAMP (synaptobrevin). • Toxicological Significance - blocks transmission- 20S- target neurotoxins, Botulinum
