  4. Study sites and sampling Group A 養殖雞 Group B 野雞 • 區域:Lublin • 樣本數量:各45隻,皆須在同一年十二月取得

  5. Laboratory procedures (實驗步驟) • 從羽毛中分離出DNA • 以QIAamp® Mini Kit在56○C培養24小時 • 以PAPD-PCR 技術增加DNA片段 pair I: 1. ABI-01 5’GTTTCGCTCC3’ 2. ABI-02 5’TGACCCTGG3’ pair II: 1. PRZ-01 5’ATGAGGTTA3’ 2. PRZ-025’ATGACGTTGA3’ (每個PCR反應總體積為25 μl,裡面包含50 ng/μl of DNA, 2.5 mMMgCl2,10 x PCR buffer(緩衝液), 2.5 mM of each dNTP, 1 U.I.ofTaq polymerase(嗜熱菌產生的耐高溫聚合酶) and 50 ng/μlprimer)

Laboratory procedures (實驗步驟) • Heat shock 1) 第一次變性:在94OC作用五分鐘 2)denaturation at 94°C for 1 min starter addition at 36°C for 2min elongation at 72°C for 1 min (46次!!!) 3) 最後再於72OC中elongation 10分鐘。 (PAPD-PCR 在MJ Research PTC-225 Peltier Thermal Cycler 中反應) • 在混有溴化乙錠的1.8%洋菜膠中,以80V電泳210分鐘 • 處理結果及數據

  7. Data analysis (數據分析) • 以DNA在電泳中分離的相片來分析 • 利用雙等位基因模式來比較每個帶(出現相同的,即得一分,若無則為零分。累計相對應之後,即可得知親緣關係。) • 在統計分析上會使用Nei’s平均基因多樣性指數(H)和Shannon指數(S)。而遺傳身份和計算遺傳距離由PopGen軟件(V. 1.31)來執行 • 基因相似樹狀圖則是利用係數和非加權對群法以及電腦的計算機程序(MEGA4)和平均數(UPGMA)來輔助求出
