  1. 研習報告與討論 蔡騰寬901607 中研院生醫所林陽生老師實驗室

  2. Interaction of TCF4 with DP103 and FHL3

  3. Abstract • Wnt signal pathway, especially beta-catenin/TCF4 complex, plays a critical role in colorectal cancer development. • By means of yeast two-hybrid assay and ECL Western blotting, DP103 and FHL3, respectively show interaction with TCF4.

  4. Introduction • Wnt signaling plays a crucial role in the etiology of colorectal cancer. • When Wnt signal is absent, the levels of cytoplasmic beta-catenin are low, and T cell factor 4 (TCF4) in the nucleus is repressed.

  5. Canonical Wnt Pathway

  6. Materials and Methods • TCF4 Cloning • Yeast Two-Hybrid Assay • DNA Sequencing • ECL Western Blotting

  7. TCF4 Cloning

  8. Yeast Two-Hybrid Assay

  9. Yeast Two-Hybrid Assay Reporter constructs

  10. Yeast Two-Hybrid Assay Yeast Colonies

  11. ECL Western Blotting

  12. Results • Transformed E. coli Is Expressible • Interaction of TCF4 Is Observable in Yeast • Interaction of TCF4 Is Detectable in Animal Cell

  13. Discussion • Benefits in Yeast Two-Hybrid System • Limitations in Yeast Two-Hybrid System • Proposed Model for TCF4, DP103 and FHL3 Expression

  14. Proposed Model for TCF4, DP103 and FHL3 Expression

  15. Conclusion • Interaction of TCF4 with DP103 and FHL3 is detectable in yeast and animal cell.
