家兔體內藥物乙醯化反應之研究. 本研究之主要目的在於探討家兔體內藥物乙醯化反應與血液中 pyruvic acid (PA) 與 α-ketoglutaric acid (α-KG) 之濃度之關系。 本實驗分二部分進行: 一、家兔乙醯化表現型之篩選 將 sulfadiazine (SDZ) 以 20 mg/kg 之劑量靜脈投予健康雄性家兔,利用高效液相層析 方法檢測血漿中 SDZ 之濃度,依 SDZ 消失速率之快慢將家兔區分為慢速或快速乙醯化 表現型 (slow or rapid acetylator phenotype) 。

  1. 家兔體內藥物乙醯化反應之研究 • 本研究之主要目的在於探討家兔體內藥物乙醯化反應與血液中pyruvic acid (PA) 與 • α-ketoglutaric acid (α-KG)之濃度之關系。 • 本實驗分二部分進行: • 一、家兔乙醯化表現型之篩選 • 將sulfadiazine (SDZ)以20 mg/kg之劑量靜脈投予健康雄性家兔,利用高效液相層析 • 方法檢測血漿中SDZ 之濃度,依SDZ 消失速率之快慢將家兔區分為慢速或快速乙醯化 • 表現型(slow or rapid acetylator phenotype)。 • 二、家兔體內PA 及a-KG濃度之測定 • 自家兔之動脈及靜脈分別采血,并將所得檢品分為血漿(plasma)及全血(whole blood • ) 處理,以高效液相層析方法檢測檢品中PA與a-KG之濃度。 • 經乙醯化表現篩選之家兔共有33只,其中有3 只為慢速乙醯化型,30只為快速乙醯化 • 型。慢性乙醯化型家兔體內PA之濃度依動脈血漿,全血;靜脈血漿,全血,分別為 • 14.60±4.89, 8.58±5.02, 12.05±4.69, 5.86±4.10 ug/ml; a-KG之濃度則分別為 • 2.27±0.57, 1.82±1.04, 2.08±0.22, 1.57±0.73 ug/ml。快速乙醯化型家兔體內 • PA之濃度依動脈血漿,全血;靜脈血漿,全血,分別為28.79±8.12, 19.03±6.99, • 27.93±7.99, 18.89±6.12 ug/ml; a-KG之濃度則分別為1.95±1.10, 1.91±0.90, • 1.80±0.85, 1.78±0.82 ug/ml。以ANOVA 檢定二種乙醯化表現型之家兔體內PA或a- • KG之濃度之差異民現無論是動脈,靜脈,血漿或全血中PA濃度於二種acetylator ph- • enotype 簡皆有顯著的差異(p<0.01 或p<0.05) 存在,a-KG則否。顯示家兔體內PA之 • 濃度與藥物乙醯化反應間具有相關性存在。
