  2. Max F. Perutz Laboratories Max F. Perutz Laboratories Georg Weitzer Kapitel 1.2: Klassifikation der Stammzellen und Stammzelllinien

  3. Max F. Perutz Laboratories Max F. Perutz Laboratories Georg Weitzer Welche Stammzelllinien gibt es? Spezies • Embryonale Stammzellen ESCs + ESLCs ~20 • Primäre Keimbahnzellen PGCs >EGCs ~5 • [Mouse, Pig, Cow, Man, Chicken] • Embryonale Teratokarzinomzellen ECCs (TCCs) ~2 • Knochenmarkstammzellen • Hämatopoetische Stammzellen HSCs 0 • Mesenchymale Stammzellen MSCs 0 • Somatische Stammzellen SSCs 0 • Darm, Haut, Hirn, Gonaden, Herz, Fett … (existieren nur in „Nischen“) 0

  4. Max F. Perutz Laboratories Max F. Perutz Laboratories Georg Weitzer Kapitel 1.3: Eigenschaften von Stammzelllinien

  5. Max F. Perutz Laboratories Max F. Perutz Laboratories Georg Weitzer Was ist eine Stammzelllinie? • Selbsterneuerungsfähigkeit (self renewal capacity) • = nicht endend wollende Teilung zu Tochterzellen (TZ) • mit identischen Potenzial und Eigenschaften wie Mutterzelle (MZ). • Derzeit Passagen (P) > 100; somatische Zellen werden spätestens bei P=80 senescent. • Klonalität, Klonierbarkeit (clonality) • = Fähigkeit als einzelne Zelle zu überleben und sich zu vermehren. • In vivo wohl immer Helfer- oder Nischenzellen dabei. • Differenzierungspotenzial (potency of differentiation) • = Anzahl der Möglichkeiten in somatische Zellen zu differenzieren Anmerkung: Eutheria sind Mammalia (Säugetiere) mit Plazenta.

  7. Max F. Perutz Laboratories Max F. Perutz Laboratories Georg Weitzer Selbsterneuerungsfähigkeit Es wird angenommen das diese durch sogenannte „Stemness Factors“ gewährleistet wird. Transkriptionsfaktoren: Oct 3/4 (Gen: Pou5f1) Nanog Sox2 Wachstumsfaktoren: IL6 Familie > JAK / STAT Signaltransduktionsweg Tgf-ß Familie > Smad Signaltransduktionsweg

  9. Max F. Perutz Laboratories Max F. Perutz Laboratories Georg Weitzer Potenzialität (in vitro + in vivo) A MZ B Stammzelle C Somatische Zellen

  10. Max F. Perutz Laboratories Georg Weitzer Potenz(ialität) von ES Zellen • ES Zellen differenzieren zu “allen” Zelltypen (~200) • Keimzellen (Eizellen und Spermien) • Leber-, Herz- Fett- Blutzellen etc. Beweis: Chimäre Mäuse Embryoid Bodies in vitro in vivo

  12. Max F. Perutz Laboratories Georg Weitzer Abstufungen der Potenzialität: Totipotenz (Omnipotenz): Oocyten, 2-8 Blastomere ? (siehe Embryoentwicklung) (Spermium ?) Pluripotenz: ESCs, EGCs, PGCs (Im Gegensatz zu den Maus ESCs, machen Menschen ESCs auch trophektodermale Zellen) Multipotenz: MSCs, HSCs ? Impotenz (Nullipotenz): somatic cells - falsch: Plastizität, Transdifferentiation, Redifferenzierung - künstlich: Reprogrammierung Takahashi and Yamanaka, 2006:Link to paper Wernig et al., 2007:Link to paper
