第七节 抗体分子的改造和基因工程. 自 1975 年 Catton 和 Milstein 等研究制造出单克隆抗体( MAb )技术以来,获得了迅速的发展和极为广泛的应用。但也确实存在不少难以克服的问题,如绝大多数的 Mab 是鼠源性的,对人有较强的免疫原性。如果用于人的治疗,不出 2 周就会产生人对鼠血清的免疫应答,患者体内存在的人抗鼠抗体( human anti-mouse antibody, HAMA ),不仅降低了疗效,严重的可引起患者出现血清病。.
