  2. what is IGF-IR? • IGF-IR( Insulin-like growth factor I receprtor) 在結構上和傳導新陳代謝訊息的胰島素接受器很相似,是一種跨膜的酪胺酸激酶接受器,它能藉由細胞膜外區域和IGF-I與IGF-II的配體結合,啟動其構形改變並向細胞膜內傳遞訊息,接著接受器會自體磷酸化數個自身細胞膜內的酪胺酸殘基,磷酸化的酪胺酸接受器便會完全活化成酪胺酸激酵素去接近並啟動下游的傳導物質,進而產生主要為細胞生長和分化現象的重要生物功用,另外,IGF-I接受器還和細胞死亡(apoptosis)以及細胞移動(motility) 作用有關

  3. About my experiment • We have known something difference between IGF-IR a and IGF-IR b • We produced two IGF-IRa promoters with different length. We put them into vectors, and used RFP (red fluorescent protein) for reporter gene. • In cell line • Microinject these vector into zebrafish embryos, and observe them to see when and where IGF-IRa express.

Method (1)用拉針條件為第一階段 Heat=726 Pull=50 Vel.=10 Time=50,第二階段 Heat=736 Pull=100 Vel.=50 Time=100的拉針器拉針,利用鑷子鑷開針口,使針口直徑在10μm以內。 (2)收取一細胞期的斑馬魚受精卵以進行顯微注射。 (3) 把卵排在排卵盤上,調整注射壓力和注射時間,使吸滿Vector的針插入受精卵,注射時注出量不要超過卵體積的1/20,避免卵的大量死亡。本實驗共分四組,分別注入CMV/RFP(+phenol red)、IGF-IRTP1443/RFP(+phenol red)、IGF-IRTP196/RFP(+phenol red)和滅菌水。 (4)每天為四組受精卵更換一至二次的水,觀察其是否有發紅色螢光的區域。為便利觀察,在24小時加0.003%PTU去黑色素,於一天的魚使用ME-222麻醉,二天以上的使用0.05%2-PE麻醉後再使用螢光顯微鏡觀察,觀察為期五天。

Discussion • 因為在7/25注射的那一批中IGF-IRPT1443/RFP有發光現象,故後續的數批顯微注射都希望能看到相同的結果,但遺憾地,沒看到一樣的結果,最可能的解釋是,7/25注射的四組未在其底盤標示,僅標示於蓋子上,在換水時不慎IGF-IRPT1443/RFP和CMV/RFP錯置,造成後續觀察上方向的誤導,且CMV/RFP一組yolk也沒有特別亮的情形發生,可以支持此一說法,但另一方面而言,預期IGF-I接受器會在快速分裂的發育部位作用,而斑馬魚應於受精後兩三天內發育最為迅速,因此在這時間點內卻沒有在實驗組看到螢光區域,有幾個可能因素,一是IGF-IRPT1443/RFP不易鑲入,使魚隻不表現螢光;二是IGF-IRPT1443/RFP的表現太弱,不易被觀測到,甚至會被魚體其他組織阻擋;三是IGF-IRPT1443的最強表現處並非在分裂發育處,而是其他階段,因此在幼體無法觀測到螢光。建議往後的實驗還是將DNA濃度提高,增加其鑲嵌率,至於其表現時期,恐怕要利用細胞培養再加以確認其最強表現其為何,再重新設定一次預期表現期。
