Respiration of Germinating Seeds Background • All living organisms need energy to survive, grow, and reproduce. • Organisms get their energy in the form of ATP (Adenosine Triphosphate), a small molecule packed with energy. • Organisms convert sugars and other macromolecules into ATP molecules through a process called cellular respiration.

Respiration of Germinating Seeds Pre Lab Set Up (Your teacher may have already completed these steps for you) • 24 hours prior to the lab: • Place 20 dry seeds in a beaker and fill the beaker with tap water. • Put the beaker in a dark location for 24 hours. The seeds will begin to germinate. • 30 minutes prior to the lab: • Place half of the germinating seeds into an ice water bath. • Allow them to cool before starting the experiment.

  9. Respiration of Germinating Seeds Safety • Use all standard laboratory safety procedures. • DO NOT allow the Carbon Dioxide Gas Sensor to get wet.

Respiration of Germinating Seeds Materials and Equipment Collect all of these materials before beginning the lab. • PASPORT Carbon Dioxide Gas Sensor • PASPORT Extension cable • Sampling bottle (included with sensor) • 1 Beaker (1000 mL) • 10 dry seeds • Ice (cubes or crushed) • Paper towels • Tap water • 10 germinating seeds at room temperature • 10 germinating seeds in ice water CO2 Gas Sensor PASPORT Extension Cable

Respiration of Germinating Seeds Setup: Dormant Seeds Q1: Why is it important to firmly insert the sensor into the bottle and create a seal? • Connect a PASPORT extension cable to the SPARK Science Learning System. Plug the CO2 Gas Sensor in the cable and then press the green button on the sensor to calibrate it. • Place 10 dormant (dry) seeds into the sampling bottle. • Put the end of the CO2 Gas Sensor into the sampling bottle. Firmly plug the end of the sampling bottle with the rubber stopper. Do not let the bottle tip over!

Place the bottle into the icy beaker so the seeds stay cold! • 3. Re-insert the sensor into the bottle. • Tap to begin collecting data. • Record data for 10 minutes, then tap to end data collection. • Remove the sensor and the chilled seeds from the sample bottle. • Rinse the sample bottle out with tap water.
