  1. Prospects for defeating aging altogether Aubrey D.N.J. de Grey, Ph.D. Chief Science Officer, SENS Foundation aubrey@sens.org

  2. SENS Foundation SENS Foundation is a US-registered charity that works to develop, promote and enable widespread access to regenerative medicine solutions to the disabilities and diseases of… AGING

  3. Who dies of aging? Death from aging is not only “natural causes” It’s anything that mainly kills older people About 150,000 people die per day worldwide Two thirds of them die of aging: 100,000/day In the USA, the proportion is over 90%

  4. Why are we doing this? Fun Not fun

  5. Structure of this talk Repair versus retardation Specifics: the seven types of damage Intracellular junk/medical bioremediation Longevity escape velocity: the concept Some evidence that LEV is realistic How you can make a difference

  6. Structure of this talk Repair versus retardation Specifics: the seven types of damage Intracellular junk/medical bioremediation Longevity escape velocity: the concept Some evidence that LEV is realistic How you can make a difference

  7. What is… • Regenerative medicine? • Aging?

  8. What is regen med? Any intervention that seeks to restore the structure of a tissue/organ to its state before it suffered damage.

  9. What is aging? Metabolism ongoingly causes “damage”. Damage eventually causes pathology.

  10. Options for intervention Gerontology Geriatrics Metabolism Damage Pathology

  11. Problem: this is metabolism

  12. If VW Bugs were built to last

  13. Maintained to last

  14. Options for intervention Gerontology Maintenance Geriatrics Metabolism Damage Pathology Claim: unlike the others, the maintenance approach may achieve a big extension of human healthy lifespan quite soon.

  15. Structure of this talk Repair versus retardation Specifics: the seven types of damage Intracellular junk/medical bioremediation Longevity escape velocity: the concept Some evidence that LEV is realistic How you can make a difference

  16. Reasons for the maintenance approach It targets initially inert intermediates (“damage”).

  17. Reasons for the maintenance approach It targets initially inert intermediates (“damage”). Damage is simpler than metabolism or pathology.

  18. This is the damage • Junk - Inside Cells • Junk - Outside Cells • Cells - Too Few • Cells - Too Many • Mutations - Chromosomes • Mutations - Mitochondria • Protein Crosslinks Seven Deadly Things No new type of damage confirmed since 1982.

  19. Giving the middle-aged 30 years of extra healthy life: Robust Human Rejuvenation

  20. Structure of this talk Repair versus retardation Specifics: the seven types of damage Intracellular junk/medical bioremediation Longevity escape velocity: the concept Some evidence that LEV is realistic How you can make a difference

  21. Giving the middle-aged 30 years of extra healthy life: Robust Human Rejuvenation

  22. Intracellular junk in the artery Endothelial Cells Lipid-engorged Lysosome Foam Cell

  23. Bioremediation: the concept Microbes, like all life, need an ecological niche. Some get it by brawn (growing very fast)… …some by brain (living off material that others can't). Any abundant, energy-rich organic material that is hard to degrade thus provides selective pressure to evolve the machinery to degrade it. That selective pressure works. Even TNT, PCBs…

  24. Xenocatabolism: the concept Graveyards are abundant in human remains, accumulate bones (which are not energy-rich), do not accumulate oxysterols, tau etc., so, should harbour microbes that degrade them… …whose catabolic enzymes could be therapeutic

  25. Environmental decontamination in vivo

  26. 7-ketocholesterol degradation a promising start 7KC over time in enrichment cultures days Biodegradation 2008; 19(6):807-813

  27. Stable isotope labelling and LC/MS reveal 7-ketocholesterol metabolites in the culture supernatant 7-ketocholesterol M = 400, M13C = 401 Hydroxylated dione ? M = 414, M13C = 415 Dione metabolite ? M = 398, M13C = 399 Culture growing on 7-ketocholesterol Culture growing on 13C-labeled 7-ketocholesterol

  28. pEGFP-COXL1: a lysosomally targeted cholesterol oxidase LAMP1 GFP Transmembrane domain and cytoplasmic tail Signal peptide Mature DS1 cholesterol oxidase

  29. COX/LAMP1 co-localizes with AO Acridine Orange EGFP Merge

  30. pEGFP-COXL1 transfected cells are protected from 7KC-induced toxicity * * * * * * *

  31. Steps to biomedical application Isolate competent strains; select by starvation. Identify the enzymes (mutagenesis, chemistry, genomics). Make lysosome-targeted transgenes; assay cell toxicity. Assay competence in vitro (more mutagenesis/selection). Construct transgenic mice; assay toxicity in vivo. Assay competence in disease mouse models. Test in humans as for lysosomal storage diseases.

  32. Structure of this talk Repair versus retardation Specifics: the seven types of damage Intracellular junk/medical bioremediation Longevity escape velocity: the concept Some evidence that LEV is realistic How you can make a difference

  33. Giving the middle-aged 30 years of extra healthy life: Robust Human Rejuvenation

  34. Reasons for the maintenance approach It targets initially inert intermediates (“damage”). Damage is simpler than metabolism or pathology. Repairing damage buys time.

  35. Progress avoids diminishing returns max Reserve frail 0 0 Age Fixing half the damage, then 3/4, then 7/8, … (a) outpaces the so-far-unfixable damage, and (b) maintains healthspan indefinitely.

  36. Longevity escape velocity (LEV) The rate at which rejuvenation therapies must improve (following the achievement of RHR) in order to outpace the accumulation of so-far-irreparable damage.

  37. Structure of this talk Repair versus retardation Specifics: the seven types of damage Intracellular junk/medical bioremediation Longevity escape velocity: the concept Some evidence that LEV is realistic How you can make a difference

  38. What rate of progress is realistic? 1903 1927 1949 1969

  39. Simulating aging (Phoenix & de Grey, AGE 2007; 29:133) Metabolism ongoingly causes “damage” and damage eventually causes pathology. so… Simulations of aging (and intervention) should simulate damage accumulation.

  40. Results: LEV is very easy Therapies double efficacy only every 42 years. Proportion dead by age N Age (years) 0 50 100 150 200 250 300 350

  41. What this means The first 1000-year-old is probably less than 20 years younger than the first 150-year-old.

  44. Support SENS Foundation SENS Foundation works to develop, promote and enable widespread access to regenerative medicine solutions to the disabilities and diseases of aging. To be successful in our mission we need your support. Time - On our website you will find details of how your skills can be used to accelerate our progress, and help us to make these audacious ideas into a reality. Money - You may also wish to make a donation. Any amount, no matter how small, helps to fund essential research, education and outreach initiatives.
