  1. Culture Media

  2. A culture medium (media, plural) is nutrient material prepared in the laboratory for the growth of bacteria, molds, and other microorganisms. The most popular and widely used substance used in microbiology laboratories is the solidifying agent agar . . Agar is a complex polysaccharide derived from seaweed (red algae)

  3. Very few microorganisms can degrade agar, so it usually remains are usually contained in test tubes or The melting point of agar is 97-100 °C and solidify at Petri dishes. 42 °C. Usually agar is used in 1.5-2% (final concentration) to solidify the liquid medium, 0.5-1% to make themedium semisolid, 5% to decrease bacterial motility (prevents swarming, proteus).

  4. Advantages of culture media • To culture microorganisms. • • For pure culture isolation. • • For storage of culture stocks. • • To observe specific biochemical reactions. • •As transport media to preserve bacteria during transportation to the laboratory.

  5. Types of culture media • A- According to physical consistencies • 1. Liquid media, broth. • 2. Solid media, Agar. • 3. Semi solid media, Gelatin

  6. B- According to the purpose of application 1. Simple media 2. Differential media. 3. Selective media . 4. Enriched media . 5. Transport media.

  7. 1- Simple media Contains the essential nutrients as source of nitrogen and carbon such as nutrientbroth, peptone water, nutrient agar. Differential media Differential media are media that contain substances that cause some bacteria to take on a different appearance from other species, allowing one species from another, e.g. MacConkey agar, mannitol alt agar, and blood agar .

  8. 3- Selective media Selective media are media that allow only certain types of organisms to grow in or on them due to the presence of inhibitory substancesor the absence of critical nutrients that make it unfavorable for most for most, but not all, organisms .the inhibitory substance may be salt (NaCl), acid, a toxic chemical (crystal violet),

  9. 4- Enriched media Certain types of bacteria need special nutrients in order to growth, simple media can be enriched by adding vitamins, yeast and salt e.g. • Blood agar. • Heated Blood agar (Chocolate agar) .

  10. 5- Transport media Simple media used for transport samples from different regions to the lab. e.g . Stuart transport medium.
