  1. Identification of Unknown Streptomyces Species Bobby Hix, Muhammad Khan, Mikayla Miller, Roger White

  2. Introduction Streptomyces sp. 5A is the organism to be used in this proposed research. Streptomyces is the largest genus of actinobacteria which are Gram-positive, filamentous bacteria that have a high GC content. Experimental question:What is the identity of the unknown Streptomyces species?

  3. Experimental Design We created growth mediums and streaked the unknown (Streptomyces sp. 5A), along with 3 reference strands (Streptomyces showdoensis, Streptomyces virudibrunneus, Streptomyces cinereoruber) onto separate plates for morphological comparison. After morphological comparison, we isolated DNA for genomic sequencing.

  4. Phylogenetic Tree

  5. Mediums/Alternate Substrates Used • TSA • Yeast Extract-Malt Extract • Peptone Yeast • Tyrosine • Glycerol-Asparagine • Oatmeal • Inorganic Salt Starch • Carbon Utilization • Tryptone-Yeast Broth • Indole Nitrate Broth • SIM • Urea • Gelatin • Citrate • 0.2% PHB w/SNC • Sucrose • Cellulose • D-xylose • I-inositol • Raffinose • D-mannitol • D-Glucose • No Carbon • Source • L-Arabinose • Rhamnose • D-Fructose

  6. Hypothesis We hypothesize that Streptomyces sp.5A is a new species.

  7. Results: Carbon Utilization- Day 7 Growth

  8. Results: Peptone Yeast Medium (15 days)

  9. Results: 0.2% PHB w/SNC Plates

  10. Results: Alternative substrates

  11. Conclusion We can accept our hypothesis because our results show that Streptomyces 5A is a different species compared to the three reference strands. Our proposed new name for this new Streptomyces species is Streptomyces nymphoides

  12. Acknowledgements Thank you, Dr. Baron, for his trust with continuing his research & help throughout the project.
