E-mail: subhasis_ceutics@yahoo.co.in Purpose: The aim of the study was to prepare Amoxicillin trihydrate microspheres with semi-synthetic and synthetic polymers for comparative evaluation of swelling factor, entrapment efficacy and in vitro release performances. Materials and Methods:Microspheres were prepared using Amoxicillin trihydrate as a model drug, Sodium carboxy methyl cellulose (NaCMC) as semi-synthetic polymer, Carbopol 940 as synthetic polymer, fused Calcium chloride and Na-alginate by inotropicgelation method. Chemical reaction between matrix forming Na-alginate polymer and divalent cation salt like Calcium chloride was utilized for development of microspheres.λmaxof Amoxicillin trihydrate was measured at pH 1.2 by JASCO V-550 double beam UV/Vis spectrophotometer. Calibration curve was prepared using above medium. Swelling study, entrapment efficacy and in vitro release study have been performed for optimization of microspheres. Fig-3:Swelling factor of the Microspheres Fig-4:In-vitro release profile of Amoxicillin microspheres in pH-1.2 (acidic buffer) Fig-1:Carbopol 940 microsphere Fig-1:Na CMC microsphere Conclusion:Sodium CMC blended microsphere is better formulation for controlled release of Amoxicillin trihydrate than Carbopol 940 with respect to swelling study, entrapment efficacy and in vitro release study. Result:Swelling factors of different formulations containing NaCMC and Carbopol 940 were 52.5% and 16.0%. Drug entrapment efficacy is also greater in case of NaCMC microsphere (55.67%) than Carbopol 940 (49.52%) respectively. Microspheres prepared with both polymers were able to release the drug constantly for 8 hr study. Sodium CMC and Carbopol 940 blended polymer showed 86.28% and 81.59% release respectively.
