Escherichia coli形成Biofilm的3種成份:

Curli :讓細胞能附著不同表面和互相黏著

(構成基因:csgG-csgC )

Colanic acid : Biofilm結構的主體

(構成基因:wcaL-wza )

Type I pili : 初始附著

(構成基因:fimB-fimH )


目的

  • 藉由去除以上三種成份的構成基因,檢視去除這些基因的影響性,和生技上的益處。


  • 利用Red-mediated markerless deletion

    技術去除三種基因,製造出一個不會生產生物膜的新菌株BD123

  • 為一種較有效率的同源重組技術

  • 具有同源序列短(40~60 bp)、重组效率高的特点.這种技術可在DNA目標分子的任意位点進行基因敲除、敲入、点突變等操作,無需使用限制性内切酶和連接脢.


結果

  • 製造出來的菌株BD123增加了重組蛋白的分泌度、抗生素的敏感度、轉型率增加。

  • 可以用此特性使BD123作為一個良好的host用於各方面的生物技術 。
