  2. Observing bacteria using the oil immersion lens: Before you focus, move the stage as high as it will go. Focus first with 10X low power Then focus with 40X high power Then add oil Then make observations with 100X oil immersion

  3. Differentiating among Gram positive bacteria Catalase test Mannitol salt agar Blood agar Endospores

  4. Catalase Test Unlike last time you performed this test, add organism to glass slide and then add H2O2.

  5. Catalase Test bubbles (gas) Distinguishes streptococci from staphylococci. Distinguishes among some Gram positive rods.

  6. Mannitol salt agar Differentiates among staphylococci

  7. Mannitol salt agar Staphylococcus ferments mannitol(yellow) Micrococcus does not ferment mannitol(pink) Differentiates among staphylococci

  8. Blood Agar Plate Reliable test Distinguishes among some streptococci and staphylococci based upon differences in hemolysis (alpha, beta, gamma)

  9. Gr + rods: Endospores present?
