  1. 嗜菌體展現胜?系統篩選技術在肺癌細胞特異性之分子診斷與造影之應用嗜菌體展現胜?系統篩選技術在肺癌細胞特異性之分子診斷與造影之應用 嗜菌體展現胜肽系統(phage display peptide system)為目前研究分子間交互作用的有利工具,本實驗分成兩部份,第一部份利用該系統對於辨認小鼠肉瘤細胞篩選出的一段胜肽鏈,結合分子造影系統,即時觀察生物活體內胜肽追蹤腫瘤的情形。經由實驗發現,將化學合成的胜肽鏈同時接上螢光物質FITC及放射線同位素131I,用核醫造影及螢光造影來偵測胜肽在體內移動的情形看出該胜肽能辨認到老鼠皮下腫瘤的位置,同時胜肽隨著時間點增加而被代謝出體外,將老鼠犧牲進一步取出各器官分別進行放射線偵測及螢光偵測來更加確定胜肽在腫瘤的位置,顯示該胜肽能辨認到腫瘤細胞。第二部份則利用該系統,針對人類非小細胞肺癌H441初步篩選出一段重複性較高的胜肽,或許可能與H441有較好的結合能力,有待往後進一步的體外體內測試。嗜菌體展現胜肽系統是個快速方便研究各種分子間關係的方式,結合上分子造影系統更能方便且清楚了解活體內的情形,有利於將來監測及診斷疾病上的重要依據。
