HYDRALAZINE之藥物動力學研究. 本研究主要目的在探討 hydralazine〔l-hydrazino-phthalazine , HP〕 投與家兔體內 後之動態:將 HP 及其代謝物 methyltriazolophthalazine(MTP 及 hydralazinepyruvic acid hydrazone(HPH) 以 0.0433mmole/kg 劑量,分別靜脈投與家兔後,利用高效液相 層析法,檢測血漿中 HP 及其代謝物的濃度,以求得各藥物動力學參數。 在投與 HP 及其代謝物後,均無法在血漿中偵測支 HP ,但投與 HP 後可見 HPH 的量迅速增

HYDRALAZINE之藥物動力學研究 • 本研究主要目的在探討hydralazine〔l-hydrazino-phthalazine,HP〕投與家兔體內 • 後之動態:將HP及其代謝物methyltriazolophthalazine(MTP及hydralazinepyruvic • acid hydrazone(HPH)以0.0433mmole/kg 劑量,分別靜脈投與家兔後,利用高效液相 • 層析法,檢測血漿中HP及其代謝物的濃度,以求得各藥物動力學參數。 • 在投與HP及其代謝物後,均無法在血漿中偵測支HP,但投與HP後可見HPH 的量迅速增 • 加,推論當HP進入體內後即很快地與血中的丙酮酸(pyruvic acid)結臺成HPH ,進而 • 被排出體外: • 比較投與HP,HPH之生成比例與投與sulfametha-zine(SMZ) 後,SMZ 在兔體內之半衰 • 期,發現兩者間有良好的線性關系(r=0.9826,p<0.01),此結果和觀察INH 代謝之結 • 果一致,顯示抱合反應與乙醯化反應間有互動關系,推論此和血中丙酮酸濃度有關。 • 另外,在投與MTP 後,其清除率及曲線下面積和SMZ 在兔體內的半衰期亦呈現良好的 • 關系性,顯示排除乙醯代謝物的能力也和個體的乙醯代謝能力有關。
